CHD2
پروتئین شمارهٔ ۲ متصلشونده به دیانای-کرومودومین-هلیکاز (انگلیسی: Chromodomain-helicase-DNA-binding protein 2) که بهاختصار «CHD2» نامیده میشود، نام آنزیمی است که در انسان توسط ژن «CHD2» کُد میشود.[۴][۵]
جهش در این ژن، سبب ایجاد انسفالوپانیهای تشنجدهنده میشود.[۶][۷][۸][۹][۱۰]
امروزه همچنین این ژن را با بروز سندرم لنوکس-گستو مرتبط میدانند.[۱۱][۱۲]
عملکرد[ویرایش]
مشخصه خانواده پروتئینهای CHD، حضور دومینهای کرومو (تعدیلگران ساختار کروماتین) و دومینهای هلیکاز/ATPase است. ژنهای CHD، احتمالاً بیان ژن را با تغییر ساختار کروماتین تغییر میدهند و بدین طریق دسترسی دستگاه رونویسی به DNA الگوی کروموزومیاش را تغییر میدهند. CHD2 فرایند سرهمبندی شدن کروماتین به آرایههای متناوب را کاتالیز کرده و ناحیه پایانه-N از CHD2 شامل کرومودومینهای پشت سرهمی است که در فعالیتهای ATPaseی و اتصال به DNA نقش خود-بازدارندگی دارد.[۱۳] برای این ژن، واریانتهای رونویسیِ پیرایش دگرسان که ایزوفورمهای متمایزی را کد میکنند نیز یافت شدهاست.[۵]
اهمیت بالینی[ویرایش]
جهشهای «دِ نووو» و حذفهای این ژن را با مواردی از حملات انسفالوپاتی مرتبط دانستهاند.[۶][۱۴][۱۵][۱۶][۱۷]
شواهد فزاینده، حملات CHD2 را به عنوان زیر-جمعیتی از سندرم لنوکس-گستو شناسایی کردهاند.[۱۸][۱۹]
